If there is a high homogeneity background, the reasons may be identified from three aspects: antibodies, experimental procedures, and buffer solution:
(1) Antibody related
The concentration of the first and second antibodies is relatively high, and the most suitable antibody dilution should be selected to optimize the dilution conditions; Alternatively, if the incubation temperature for the first antibody is too high, it is recommended to incubate overnight at 4 ℃.
Antibody inactivation due to long storage time or poor storage conditions: It is advisable to choose a new antibody.
